The casualties were sustained in a firefight with Taliban forces north of the city of Gerishk in the restive Helmand province, the Army Operational Command statement said.
Since 2002, 14 Danish soldiers have been killed in Afghanistan, including two earlier this month.
Prime Minister Anders Fogh Rasmussen and Defence Minister Soren Gade expressed their condolences as did Major General Poul Kiaerskou of the Army Operational Command.
Rasmussen said it was important to "fully support" the Danish forces based in Afghanistan.
Denmark has a 600-strong force in Afghanistan.
